If the visa documents are submitted reflecting all the nuances of the trip (tickets and a voucher for accommodation in Romania) - no problems will be neither with the opening of a visa, nor with entry into the territory of Spain, nor with the re-opening of a visa. Problems arise when it follows from the documents that the first country of entry is Spain, but this is not true.
